敗血症の新しい治療法を発見!
私達は、カイオムバイオサイエンス社と共同で、選択的抗セマフォリン3Aモノクローナル抗体の開発に成功しました。セマフォリン3Aは神経回路形成に重要な役割を担う蛋白質であり、神経成長末端に形成される成長円錐を退縮させ、突起の伸長を抑制する作用があります。現在、セマフォリン3Aは神経系のみならず、心血管系、免疫系、骨代謝に関わる重要な分子であることが分っており、がんや免疫疾患との関わりも示唆されています。今回、その作製に成功したセマフォリン3A抗体は、ニワトリ後根神経節細胞の成長円錐を退縮するセマフォリン3Aの効果を阻害する活性を示すことに加え、敗血症モデルマウスの生存率を高める効果を持つことが明らかになりました。この抗体の投与は、敗血症増悪因子の一つである血中のプラスミノーゲンアクチベーター1を抑制します。様々な感染症にともなう敗血症の致死率は依然として高く、有効な治療法も乏しいなか、今回の成果は、敗血症にともなう著しいサイトカイン過剰産生、サイトカインストームを抑制するという全く新しい治療法の開発に繋がるものとして注目されます。 (詳細はこちらをご確認下さい)

神経ガイド分子セマフォリン3Aの新たな作用メカニズムを解明
- 神経回路が形成される時に、神経ガイド分子とよばれるタンパク質が神経突起の伸長をコントロールします。神経ガイド分子のうち、神経突起を遠ざける反発因子の一つであるセマフォリン3A (SemaA)は、神経の伸びを阻止します。私たちは、アクチン結合蛋白質の一つであるフィラミンA(Filamin-A)が、CRMP1(細胞内蛋白質の一つ)と結合してアクチン骨格をコントロールしている事実を発見しました。私たちは、このとき、これらの分子複合体の構造が大きく変化していることを原子間顕微鏡を用いて明らかにしました。この成果を得るにあたり、科学研究費基盤研究(C)、文部科学省「イノベーションシステム整備事業」、グローバルCOE「情報通信による医工学融合イノベーション創生」により支援を受けました。ここに謝辞を申し上げます。

神経回路形成の新しいメカニズムを発見
グルタミン酸は私達の脳で学習や記憶を司る重要な働きをしています。グルタミン酸が働くためにはそれと結合して脳内の情報を伝達するグルタミン酸受容体が、神経細胞の一定の場所に存在していることが必要です。私達は、神経の伸長方向のコントロールに重要な役割をするセマフォリン3Aというタンパク質が、グルタミン酸受容体の神経細胞の細胞体や樹状突起における場所を、今までにない新しい仕組みで神経突起の末端の遠隔からコントロールするというメカニズムを発見しました。

私達は、ドパミンの前駆体であるドーパが神経伝達物質であることを提起してきました。この論文は、私達が長年検討してきたドーパの作用が、Gpr143、別名OA1 (ocular albinism 1)と呼ばれるG蛋白質連関型受容体を介して発現することを世界に先駆けて明かにすることに成功しました。この発見は、ドーパ神経伝達物質仮説をさらに支持するものであり、今後、全く新たな発見、創薬研究の基礎となるものと考えられます。
